摘要:
双光子激发受激发射损耗(two-photon excitation stimulated emission depletion,2PE-STED)显微镜是一种新型的非线性超高分辨荧光显微镜.2PE-STED显微镜在突破衍射极限 基础上,具有穿透深度深、图像信噪比高等优势.相比于其他超高分辨显微镜,2PE-STED显微镜可应用于具有高散射特性的生物活体组织的超高分辨层析成像.本文详细介绍了2PE-STED显微技术的基本原理、不同实现形式及其在生物研究中的应用,并展望了其未来的发展趋势.
Abstract:
Two-photon excitation stimulated emission depletion (2PE-STED) microsoope is a new kind of nonlinear super-resolved fluorescence microscope.2PE-STED microscopy can provide subdiffraction resolution with much deeper penetration depth and higher signal-to-noise ratio of image. Compared with other super-resolved microscopes, 2PE-STED microscope allows three-dimensional super-resolved imaging of thick living tissue. This review presents the principle, different system setups, and application of 2PE-STED microscopy and discusses its development in the future.
